Abstract

Objective To investigate the effects of picropodophyllin (PPP) on philadelphia chromosome positive (Ph+) acute lymphoblastic leukemia (Ph+ ALL) Sup-B15 cells and its underlying mechanisms.Methods MTS assay was used to analyze the effect of PPP and imatinib mesylate (IM) on the proliferation of Sup-B15 cells.The cell cycle distribution after PPP and IM treatment of Sup-B15 cells were determined by flow cytometry.The effect of PPP on the expression of proliferation and cell cycle-associated proteins,and the phosphorylation and expression of IGF1R was determind by Western blot.Results PPP and IM inhibited the proliferation of Sup-B15 cells in a dose-and time-dependent manner,and the inhibitory effect of PPP was stronger than IM at the same doses.Cell cycle analysis showed that PPP treatment for 12 and 24 h resulted in significant accumulation at the G2/M phase and decreased at S phase of Sup-B15 cells.PPP treatment increased p53 and cyclin B1 protein expression,and reduced p21 and c-myc protein expression,but had no obvious effect on phosphorylation and total protein levels of IGF1R.Conclusion PPP can effectively inhibit Sup-B15 cell proliferation and induce G2/M cell cycle arrest,which is associated with upregulation of p53 and cyclin B1 expression and inhibition of p21 and c-myc expression.
