摘要

Abstract

Objective To study the influence that icariin(ICA)exerts on the levels of ERα,ERβ and GR mRNA on PBMC in patients with multiple sclerosis (MS) and the relationship between dose and efficacy.Methods PBMC was isolated and extracted from MS patients and healthy volunteers,both of them were divided into 8 subgroups respectively including the blank control group,the estrogen group,three different doses of ICA groups and three correspondingly doses of blank serums control groups.The expression of ERα,ERβ and GR mRNA were detected by real-time quantitative PCR after all subgroups were cultured and centrifuged.Results Compared with the control group respectively,ERα mRNA expressions in ICA groups of high-,middle-doses and estrogen group of MS patients were significantly increased,and ERβ mRNA expressions were significantly increased in ICA groups of high-dose and estrogn groups of MS patients.There was no significant difference between MS patients and healthy people on levels of GR mRNA expression compared with the control groups.Compared with the healthy people group respectively,there were significant differences on ERα and ERβ mRNA expressions in MS patients groups with high-doses of ICA.Conclusion ICA has estrogenic effect and increases expressions of ERα and ERβ mRNA on PBMC in MS patients,but it has no effects on expressions of GR mRNA on PBMC in patients with MS.
