Abstract

Objective: To investigate the effect of microRNA-3619-5p (miR-3619-5p) overexpression on the up-regulation of p21 gene in ovarian cancer cell lines A2780 and SKOV3 and its effect on ovarian cancer cell growth. Methods:miR-3619-5p mimics (experimental group) or dsControl (control group) were transiently transfected into ovarian cancer cell lines A2780 and SKOV3 using Lipofectamine 3000 respectively. The expression of p21, CDK4 and Cyclin D1 mRNA was detected by real-time quantitative polymerase chain reaction (qRT-PCR). The expression of p21, CDK4 and Cyclin D1 was detected by Western blot. Flow cytometry was used to detect the cell cycle distribution and cell apoptosis in the control and experimental groups. EdU proliferation assay and colony formation assay were conducted to analyze the proliferation ability of transfected cells. Results:Compared with dsControl group, the expression of p21 mRNA, CDK4 mRNA and Cyclin D1 mRNA in cell lines transfected with miR-3619-5p were significantly increased (P<0.01). Western blot results were consistent with qRT-PCR results. Compared with the dsControl group, the percentage of cells in S phase and G 2/M phase decreased significantly after transfection of miR-3619-5p, the proportion of cells in G0/G1 phase was significantly increased, and the apoptosis rate was significantly increased. The proliferation ability of ovarian cancer cells transfected with miR-3619-5p was significantly lower than that of the dsControl group, as demonstrated by the EdU proliferation assay and colony-forming assay (P<0.05). Conclusions:miR-3619-5p can significantly inhibit the growth of ovarian cancer cells by activating the expression of p21 protein in ovarian cancer cells.
