Correlation of Slug expression in esophageal cancer tissue with the epithelial-mesenchymal transition and invasion of cancer cells

Abstract
Objective: To study the correlation of Slug expression in esophageal cancer tissue with the epithelial-mesenchymal transition and invasion of cancer cells.Methods: A total of 78 patients with esophageal cancer who accepted radical operation in our hospital between June 2013 and June 2016 were selected, and the esophageal cancer tissue and normal tissue specimens adjacent to carcinoma were collected during operation.Fluorescence quantitative PCR method was used to determine the Slug gene and invasion gene mRNA expression in sample tissue, western-blot method was used to determine epithelial-mesenchymal transition-related index protein expression, and Pearson test was used to evaluate the correlation of Slug expression in esophageal cancer tissue with the epithelial-mesenchymal transition and invasion of cancer cells.Results: Slug mRNA expression in esophageal cancer tissue was significantly higher than that in tissue adjacent to carcinoma (P<0.05).Epithelial-mesenchymal transition marker E-cadherin protein expression in esophageal cancer tissue was lower than that in tissue adjacent to carcinoma while Vimentin and N-cadherin protein expression were higher than those in tissue adjacent to carcinoma, and pro-invasion genes Ezrin, FAT10, Survivin, MCP-1 and RhoA mRNA expression were higher than those in tissue adjacent to carcinoma while anti-invasion genes DEC1, PTEN and p53 mRNA expression were lower than those in tissue adjacent to carcinoma (all P<0.05).Slug mRNA expression in esophageal cancer tissue was negatively correlated with epithelial-mesenchymal transition marker E-cadherin protein expression, positively correlated with Vimentin and N-cadherin protein expression, positively correlated with pro-invasion genes Ezrin, FAT10, Survivin, MCP-1 and RhoA mRNA expression, and negatively correlated with anti-invasion genes DEC1, PTEN and p53 mRNA expression.Conclusions: Slug expression in esophageal cancer tissue is higher than that in tissue adjacent to carcinoma, and Slug expression is directly correlated with the epithelial-mesenchymal transition and invasion of cancer cells.关键词
