Effects of exogenous dextranase and sodium fluoride on biofilm formation of Streptococcus mutans in different sucrose concentration medium

Abstract
Objective:To investigate the effects of exogenous dextranase(Dex)and sodium fluoride(NaF)in different sucrose concentration medium on biofilm formation of Streptococcus mutans(S.Mutans).Methods:The model of S.Mutans biofilmin in vitro was established.The pH value of the medium with different sucrose concentration during the biofilm formation was detected by pH electrode.Crystal violet staining was used to observe the effects of Dex and NaF on biofilm formation.Scanning electron microscope was applied to detect the inhibitory effect of Dex and NaF on biofilm formation.Results:In different sucrose concentration medium,Dex had no effect on the pH value,but NaF could increase the pH value.The pH values of the Dex+NaF groups were higher than the control groups(P<0.05),and significant difference was existed among Dex+NaF groups with different sucrose concentration(P<0.05) except the 3% and 4% groups.The mass of biofilm in all treated groups decreased when compared with the control groups,and Dex and NaF had synergistic inhibitory effect on the biofilm formation.There were statistically significant differences in biofilm formation between Dex+NaF groups and control groups(P<0.05).When simultaneously treated with Dex and NaF,the formation of biofilm decreased when the sucrose concentration increased.Conclusion:Dex and NaF had inhibitory effect on the biofilm formation of S.Mutans,and they had synergistic effect when used together.Sucrose concentration could also affect the biofilm formation of S.Mutans.关键词
