CTGF up-regulates the osteogenic differentiation of periodontal ligament cells induced by BMP-2

Abstract
Objective:To investigate the function of CTGF on biological effects of PDLCs induced by BMP-2.Methods:PDLCs derived from fragment of tissue were cultured in three groups,including blank group(general medium),control group(with 100 ng/mL BMP-2),experimental group(with 100 ng/mL BMP-2+50 ng/mL CTGF).Proliferation of cells in each group was measured by CCK-8 after 1,3,5 days.Activity of ALP in PDLCs was measured after cultured 5 days.The mRNA of Runx2,ALP,BSP in each group was tested by qRT-PCR.Results:The quantity of cells in experimental group was higher than that in blank group and control group after 3-day culturing,which was significant higher after 5-day culturing(P<0.05).The activity of ALP in experimental group was stronger than that in blank group and control group(P<0.05).Moreover,mRNA of experimental group showed significantly higher than that of blank group and control group(P<0.05).Conclusion:CTGF up-regulates the osteogenic differentiation of periodontal ligament cells induced by BMP-2.关键词
