二甲双胍对去势大鼠胫骨生物力学影响的研究
Effect of metformin on biomechanics of the tibiae of postmenopausal osteoporotic rats

Abstract
Objective:The purpose of this study was to investigate the effect of metformin on bone mineral density and biomechanics in the tibiae of ovariectomized rats.Methods:Three months after either a sham surgery or bilateral ovariectomy,40 female Sprague-Dawley rats were assigned into 4 groups:Sham group,OVX group,OVX+metformin(50 mg/kg/d)group and OVX+metformin(100 mg/kg/d)group.After 3 months of gastric feeding with or without metformin,tibiae were collected for dual energy X-ray absorptiometry and three-point bending test.Results:Compared with Sham group,bilateral ovariectomy induced a decline in bone mineral density.This parameter was higher when using of metformin than OVX group,but was also lower than Sham group.The maximum load and elastic modulus were obviously reduced in OVX group than Sham group(P<0.05).The parameters got higher when using of metformin,meanwhile it was dose-dependent.Conclusion:Metformin may increase both bone mineral density and the biomechanics in ovariectomized rats.关键词
