竹节香附素A通过下调长链非编码RNA HOTAIR表达抑制胃癌细胞增殖的研究

Abstract
Objective:To examine the expression levels of long non-coding RNA HOTAIR in gastric cancer cells.Examining if Raddeanin A inhibits gastric tumor growth through regulating the expression level of lncRNA HOTAIR.Methods:Real-time quantitative PCR was applied to detect the expression of HOTAIR in normal epithelial cell line GES-1 and gastric cancer MGC-803,SGC-7901,HGC-27 cell lines.The specific small interfering RNA for HOTAIR (si-HOTAIR group),negative control sequence (si-NC group),or si-HOTAIR with Raddeanin A group were transfected into MGC-803 cells.The MTT assay and clone formation experiment were employed to detect the proliferation of two groups.Results:The levels of HOTAIR was high ingastric cancer cells compared with gastric epithelial cells.Compared with si-NC group,there were lower HOTAIR level,cell activity and cell clone number in si-HOTAIR group,Raddeanin A group and si-HOTAIR with Raddeanin A group with statistical significance (P < 0.05).Compared with si-HOTAIR group,there was no statistical significance in HOTAIR expression in Raddeanin A group (P > 0.05),but there was lower HOTAIR expression in si-HOTAIR with Raddeanin A group with statistical significance (P < 0.05).Conclusion:There is high expression of HOTAIR in gastric cancer cells.Dow-regulation HOTAIR expression can inhibit gastric cancer cell proliferation.Raddeanin A can inhibit gastric cancer cell proliferation through regulating the expression level of HOTAIR.And there is synergistic action of Raddeanin A and si-RNA in inhibiting gastric cancer cell proliferation.关键词
