健脾调肠方对溃疡性结肠炎大鼠NF-kB表达影响的实验研究
Expermental Study on the Effect of Jianpi Tiaochang Recipe for the Expression of NF-kB in Ulcerative Colitis Tissue of Rats
摘要
Abstract
Objective:To explore the effect of Jianpi Tiaochang Recipe on ulcerative colitis (UC) in rats.Methods:SD rats were randomly divided into five groups,controlled group,Jianpi Tiaochang Recipe treated group at low (14.13g/kg),medium (28.23g/kg) and high (56.52g/kg) dosage group individually (n =5).The model of UC was induced by enema with 5％ 2,4,6-trinitrobenzene sulfonic acid (TNBS) /ethanol.After seven days of enema,the rats were randomly divided into different groups as above.The controlled group was given saline according to the body weight of rats.The treated group accepted enema at different dosage of Jianpi Tiaochang Recipe for 14 days.We observed the changes of colonic tissue of rats by HE staining.The expression of NF-kB and NF-kB mRNA were detected by immunohistochemistry and RT-PCR.Results:Ulcer pathology was observed in UC group and a large number of inflammatory cell infiltrated in submucosal mucosal of intestinal.Injury of intestinal mucosa was attenuated in the treated group.Compared with the controlled group,the expression of NF-kB in colon tissue were significantly increased in the model rats (P ＜ 0.05).NF-kB expression in Jianpi Tiaochang Recipe group was decreased (P ＜0.05).Conclusion:Jianpi Tiaochang Recipe can alleviate the inflammation of intestinal and its mechanism may relate to its regulation of the expression of NF-kB.关键词
