Regulation effect of long non-coding RNA HOXC-AS1 on proliferation, migration, and tumorigenesis of gastric cancer cells

Abstract
Objective To explore the regulation effect of long non-coding RNA (lncRNA) HOXC-AS1 on proliferation, migration, and tumorigenesis of gastric cancer cells.Methods HOXC-AS1 in gastric cancer cell line BGC823, SGC7901 cells and normal gastric mucosal cell line GES-1 was detected by real-time quantitative PCR.The BGC823 and SGC7901 cells of gastric cancer were divided into HOXC-AS1 small interfering RNA (siRNA) group, siRNA control sequence group, HOXC-AS1 overexpression group, and empty plasmid group.HOXC-AS1 siRNA group was transfected with HOXC-AS1 siRNA, siRNA control sequence group was transfected with siRNA control sequence, HOXC-AS1 overexpression group was transfected with HOXC-AS1 overexpression plasmid, and empty plasmid group was transfected with empty plasmid.After 24-hour culture, the cell proliferation ability of each group was observed by MTT and clonogenic assay, Transwell test and cell scratch test were used to observe the cell migration ability of each group (the result was expressed by the number of penetrating cells and the rate of scratch healing).Twenty nude mice of 5 weeks old were randomly divided into transplantation tumor group and control group with 10 in each group.The mice in the two groups were separately injected with BGC823 cells (1.5×106) from the HOXC-AS1 siRNA group and siRNA control sequence group after 24-hour transfection.The mice were killed and tumor volume was measured after 16 d.Results The expression of HOXC-AS1 in BGC823 and SGC7901 cells was 4.58±0.62 and 2.89±0.25 times as high as that of GES-1 cells, respectively.The relative expression of HOXC-AS1 in both BGC823 and SGC7901 cells was higher than that in GES-1 cells (P<0.05).The OD490, the number of clones, the number of penetrating cells and wound healing rates of HOXC-AS1 siRNA group in BGC823 and SGC7901 cells were lower than those of the control group (all P<0.05).The OD490 value, the formation of the clone number, the number of penetrating cells, and wound healing rate of HOXC-AS1 overexpression group were higher than those of the empty plasmid group (all P<0.05).The tumor volume of transplantation tumor group and control group were (353.93±147.85) and (706.24±253.25)mm3, respectively, and the weight was (0.30±0.13) and (0.75±0.11) mg, respectively.The volume and weight of tumor in transplantation tumor group were higher than those in the control group (all P<0.05).Conclusion The expression of HOXC-AS1 increases in gastric cancer cells, and LncRNA HOXC-AS1 can promote the proliferation, migration, and tumorigenesis of gastric cancer cells.关键词
