红景天苷对野百合碱诱导肺动脉高压大鼠RhoA/Rho激酶信号通路的影响
Effect of Salidroside on the signal pathway of RhoA/Rho kinase in monocrotaline-induced pulmonary hypertension in rats

Abstract
Objective: To investigate the effect of salidroside on RhoA/Rho kinase signaling pathway on lung tissue pathology in rats with pulmonary hypertension.Methods: The thirty 6-8 week old male SD rats were randomly divided to 5 groups: normal control group, model control group, low concentration group, middle concentration group, the high concentration group.In addition to the normal control group, the other groups were injected with 60 mg/kg of monocrotaline.in 2-28 day, the rats in low concentration group, medium concentration group and high concentration group were injected with salidroside at 50, 100, 200 mg/kg/d, respectively, the control group rats were injected with normal saline.At the end of the experiment, the right ventricular systolic pressure (RVSP) and the right ventricular hypertrophy index were detected in each group.The pathological changes of lung tissue were detected by HE staining in rats.The mRNA expression level of ROCK1 and RhoA in lung tissue was detected by RT-PCR.The protein expression level of ROCK1,RhoA and and phosphorylation of MYPT1 in lung tissue was detected by Western blot.Results: Compared with the normal control group, in model control group rats,the RVSP and right ventricular hypertrophy index increased significantly, the RhoA and Rock1 mRNA in lung tissue was significantly up-regulated, the protein expression of RhoA,Rock1, and p-MYPT1 were significantly up-regulated.After treatment with different concentrations of salidroside, the changes of RVSP and right ventricular hypertrophy index were significantly lower in the high concentration group.The expression of RhoA and ROCK1 mRNA in lung tissue was gradually down regulated, and the expression and phosphorylation of RhoA,ROCK1, p-MYPT1 were gradually down regulated, which was concentration dependent, and the change of medium and high concentration groups was significant.Conclusion: A certain dose of salidroside can alleviate the pulmonary hypertension induced by monocrotaline, which may be related to the inhibition of the RhoA/Rho kinase signaling pathway.关键词
