CDA基因79位点多态性预测吉西他滨治疗晚期难治性恶性肿瘤的疗效
Prognostic value of CDA gene 79 locus genetic polymorphisms in the efficacy of gemcitabine chemotherapy in treatment of advanced refractory solid tumors

Abstract
Objective To investigate the association between CDA gene 79 locus genetic polymorphisms and the efficacy of gemcitabine-based chemotherapy in treating patients with advanced refractory solid tumors.Methods Fifty patients with advanced refractory solid tumors received gemcitabine-based chemotherapy(1 000 mg/m2,intravenous drip,dl,d8,every 3 weeks).Efficacy was evaluated after treatment.Patients' DNA was extracted from peripheral blood before chemotherapy,and CDA gene 79 locus genetic polymorphisms were detected by amplifying the gene fragments using PCR and direct sequencing.The association between the CDA gene 79 locus genetic polymorphisms and gemcitabine efficacy was analyzed.Results A total of 47 patients were studied.One case had complete response,12 cases had partial response,27 cases had stable disease,and 7 cases had progression disease.The clinical relief rates were 27.7%.Two different genotypes of CDA gene 79 locus were detected:wild-type genotype A/A (40 cases,85.1%) and mutant genotype A/C (7 cases,14.9%).CDA79 genetic polymorphisms were found to be associated with clinical efficacy (P < 0.05),but not with age,gender or PS scores.Moreover,patients with CDA79 A/C tended to have higher rates of disease remission than those with CDA79 A/A (71.4% vs 20.0%,P < 0.05).Conclusions CDA gene 79 locus genetic polymorphisms can be used to predict the response to gemcitabine chemotherapy of patients with advanced refractory solid tumors.Patients who has CDA gene 79 locus mutant genotype A/C are more sensitive to the chemotherapy than patients with CDA gene 79 locus wild-type genotype A/A.关键词
