Effect of low-molecular weight heparin on invasion of in vitro cytotrophoblast cells incubated in 2% and 20% oxygen tention respectively

Abstract
Objective:To investigate the effect of low-molecular weight heparin (LMWH) on invasion of human primary first trimester cytotrophoblast cells incubated in 2% and 20% oxygen tention respectively.Methods:Cytotrophoblast cells were obtained by trypsin-collagenase digestion method from early gestational villous tissue (8~10 weeks) and incubated in 2% and 20% O2.Invasion of cytotrophoblast cells was evaluated following exposure to LMWH of different concentrations (0,0.1,5,10IU/ml).The expression of HIF-1α was detected by real-time quantitative RT-PCR method.The expressions and secretions of MMP-2,MMP-9,TIMP-2,TIMP-3 were examined by real-time quantitative RT-PCR method and ELISA assay.Results:The invasive ability of cytotrophoblast cells in early pregnancy was enhanced with increasing concentrations of LMWH (0.1,5IU/ml) when being incubated in 2% O2,but an opposite effect at the concentration of 10IU/ml LMWH.The expression of HIF-1α increased at the lower concentration of LMWH (0.1,5IU/ml),and then decreased at the higher concentration of LMWH(10IU/ml).Further data showed that the expression and secretion of MMP-2 were dominant,whereas the amount of MMP-9 was negligible.And we observed the similar changes of the expressions of MMP-2,TIMP-2 and TIMP-3 like that of HIF-1α.The invasion of cytotrophoblast cells,incubated in 20% O2,was not enhanced,and the expression levels of HIF-1α,MMP-2,TIMP-2,TIMP-3 were not significantly changed.Conclusion:LMWH can enhance the invasion of cytotrophoblast cells induced by hypoxia,which might be related with increase of the gene expression such as HIF-1α,MMP-2.关键词
