Acute Quinpirole Treatment Inhibits Sensory Motor Gating of C57BL/6J Mice

Abstract
Objective To investigate the effects of quinpirole after acute treatment on acoustic startle reflex (ASR) and sensory motor gating in C57BL/6J mice.Methods 24 mice were randomly divided into three groups,the control group (n =8),the low-dosage group (n=8),and the high-dosage group (n=8).The mice in the low-dosage group and high-dosage group were given quinpirole 1 mg/kg and 3mg/kg injection,respectively.The control group was treated with equal volume of saline.All mice received ASR and pre-pulse inhibit (PPI) tests by Startle reflex system before and after drug administration.Results The PPI rate was about 50% after the mouse was measured ASR and PPI test.The maximum startle magnitude (0.0707±0.0013) N and PPI rate (42.73 ± 7.13)% were significantly lower than those before saline injection [(0.0813±0.0047) N,(39.53±4.02) %] (P<0.05).The maximum startle magnitude (0.0654±0.0093) N in the low dose experimental group was lower than that in the control group (0.0813±0.0047) N and the control group (0.0823±0.0074)N (P<0.05),and its %PPI (32.66±6.24)% were lower than those in the control group (49.58±5.03)%,but there was no significant difference between each group (P > 0.05).The maximum startle magnitude (0.0307 ± 0.0026)N and % PPI (14.48±6.14)% which in the high dose group were significantly lower than those in the baseline group (0.0864±0.0143) N and (45.24± 10.11) %,compared with the control group (P<0.05).Conclusion Acute injection with high doses of quinpirole reduced the prepulse inhibition of auditory startle reflex,and injured the sensory motor gating function in C57BL/6J mice,probably via dysregulation of PPI in the limbic system.关键词
