雷帕霉素对PC12细胞氧糖剥夺损伤的保护作用

Abstract
Objective To investigate the effect of rapamycin on PC12 cells injured by oxygen and glucose deprivation.Methods The PC12 cells which in the logarithmic phase were randomly divided into control,OGD,vehicle,1,10,100,500 nmol rapamycin groups.Rapamycin was added to the medium directly at the start of OGD.We observed the morphology changes of PC12 cells by inverted microscope after OGD,evaluated the survival rate of PC12 cells challenged with the MTT assay.Results Compared with the control group,the PC12 cells activity of OGD group was significantly decreased (P<0.05),compared with the vehicle group,the cell survival rates of 1,10,100,500 nmol rapamycin groups were increased in a varying degrees,and cell survival rates of the 10,100,500 nmol have statistically significant (P<0.05).Conclusion Rapamycin has a protective effect in OGD injury of PC12 cells.关键词
