应用iTRAQ蛋白质组学方法筛选宫颈癌及癌前病变差异蛋白
摘要
Abstract
Objective:To explore and optimize the method of labeled relative and absolute quantification (iTRAQ) and separation of proteome in human high grade cervical intraepithelial neoplasm and cervical cancer tissue.Methods: By optimizing the trypsin and label of proteins in human intraepithelial neoplasm and cervical cancer tissue, parameters on pH-RPseparationand RPLC-MS on second dimension, to determine the method for screen and identification of proteins.Results: The proteins were successfully extracted from human ovarian cancer ascites and were separated by DIGE with higher resolution and better reproducibility.Conclusion:iTRAQ technique is a suitable approach to study the proteome in human intraepithelial neoplasm and cervical cancer tissue.关键词
