Abstract
AIM To develop a joint gradient elution HPLC wavelength switching combined with gradient elution for determination of the content of swertiamain,mangiferin,isoorientin,swertianolin,hyperoside,isoquercitrin,isochlorogenic acid C (4,5') and isorhamnetin-3-O-β-D-glucoside in Yugan tablet simultaneously.METHODS The Kromasil C tschromatographic column (250 mm× 4.6 mm,5.0 μm) was adopted;the mobile phase was acetonitrile (A)-0.04％ phosphoric acid solution (B) with gradient elution.The flow rate was 0.9 mL ·min-1,the column temperature was set at 30℃,and the sample quantity was 10 μ,L.The detection wavelength was 254 nm for swertiamain,mangiferin,isoorientin and swertianolin,345 nm for hyperoside,isoquercitrin,isochlorogenic acid C(4,5') and isorhamnetin-3-O-β-D-glucoside respectively.RESULTS The contents of swertiamain,mangiferin,isoorientin,swertianolin,hyperoside,isoquercitrin,isochlorogenic acid C (4,5') and isorhamnetin-3-O-β-D-glucosidewere 10.55-211.0 mg· L-1(r=0.999 8),14.76-295.2 mg· L-1(r=0.999 9),6.790-135.8 mg· L-1(r=0.9993),9.080-181.6 mg· L-1(r=0.999 7),4.360-87.20 mg· L-1(r=0.9999),2.320-46.40mg· L-1(r=0.9999),13.69-273.8 mg· L-1(r=0.9995) and 2.350-47.00mg· L-1(r =0.999 4) respectively within the scope of concentration and peak area had a good linear relationship.The average recoveries and the corresponding RSD were 97.97％ (1.49％),99.88％ (0.81％),98.79％ (1.10％),97.12％ (0.84％),98.21％(1.61％),100.02％ (0.78％),98.96％ (1.79％) and 99.35％ (1.52％),respectively.CONCLUSION Simultaneous determination of eight components in Yugan tablet joint gradient elution HPLC wavelength switching method is established.The method is high sensitivity,high accuracy and good specificity,and can be applied to the quality control of Yugan tablet.关键词
