Abstract

In order to explore the microbial resources for controlling rice blast,caused by Magnaporthe oryzae,527 strains were isolated from healthy rice plants of the susceptible cultivar Xiangzaoxian 24 by the method of plate dilution.60 strains with antagonistic activity against the blast fungus were obtained.The strain YN145 showed the strongest inhibition activity on mycelium growth with the rate of 84.31%.Based on the morphological analysis,physiological and biochemical characteristics,and 16S rDNA phylogeny,the strain YN145 was identified as Bacillus subtilis.Biological activity test showed that YN145 still had the inhibition rate of more than 80% on mycelium growth after 25 generations of subcultures.Fermentation supematant of the strain could inhibit the mycelium growth by the rate of 91.50%,92.35%,91.78%,90.93% and 86.97%,respectively,after treated at 40,60,80,100 and 120 ℃ for 30 min.Meanwhile,the fermentation filtrate could inhibit the mycelium growth by the rate of 91.50%,92.92%,92.92%,92.35% and 92.63%,respectively,after treated by pH 3,5,7,9 or 11 for 30 min.The results suggested that antifungal substance of YN145 may be resistant to high temperatures and anti-acid and alkali.Thus,the strain YN145 is worthy for further research and development as rice blast bio-control agents.
