Effects of Lactobacillus amylovorus on rotavirus receptor GM3 and its synthetase mRNA

Abstract
To investigate the possible mechanisms of Lactobacillus amylovorus against porcine rotavirus(PRV),3-day-old clean grade Kunming mice were fed with L.amylovorus for 4 days,and the rotavirus was inoculated on the 4th day of the experiment.One of the major receptors of porcine rotavirus (Ganglioside) GM3 was quantitatively detected by ELISA,and the mRNArelative contents of UDP-glucose ceramide glucosyltransferase(UGCG) and ST3 beta-galactoside alpha-2,3-sialyltransferase 5(St3ga15)in each sample were determined by real-time fluorescence quantitative PCR.The results showed that the content of GM3 in the L.amylovorus group was significantly higher than that in the normal group and the virus group on day 5,9 and ll(P<0.05),and the relative expression of MusSt3gal5 mRNA in the L.amylovorus group was significantly lower than that in the normal group onday5,7,9 and 11(P<0.05).The relaive expression of MusSt3gal5mRNAinthestarch group was significantlylowerthanthatinthevirus group on day 7,9 and ll(P<0.05).The relative expression of MusUGCG mRNA in the L.amylovorus group was significantly lower than that in thenormal group and the virus group on day 7,9 and 11(P<0.05).The results showed that L.amylovoruscould promote the synthesis of ganglioside GM3 in the jejunum and inhibit the expression of MusSt3gal5 and MusUGCGmRNA in the jejunum in suckling mice after infection with rotavirus.The study offers help to further study the effects of L.amylovorus against rotavirus mechanisms.关键词
