摘要

Abstract

To elucidate the mechanism of zearalenone(ZEA) on male reproductive toxicity,we investigated the effects of ZEA on the autophagy and related signaling pathways in sertoli cells.TM4 cells were used as materials treated with 0(control group),0.1,1,10,20,30 μmol/L ZEA respectively for 24 h.With the help of the CCK-8,1aser confocal microscopy,transmission electron microscopy and West ern blot technique,the cell viability,autophagy,effects of the MAPK signaling pathway and the relationship between MAPK signal pathway and the autophagy of TM4 were investigated.The immunofluorescence results showed that with the increasing concentration of ZEA,the number of the LC3 accumulation in TM4 cells significantly increased and the number of acidicvesicles also increased.The Western-blot results showed that the expression of autophagy related protein such as LC3 Ⅱ,Beclin-1,ATG5,ATG7 was significantly increased,while the expression of P62 was significantly decreased(P<0.05 or P<0.01).The phosphorylation level of the ERK1/2 in the pathway of KAPK signaling increased significantly(P< 0.05 or P<0.01).After adding the U0126 (ERK phosphorylation inhibitor),the level of ZEA induced au tophagy in TM4 decreased significantly (P<0.05 or P<0.01).The results showed that zearalenone could induce autophagy of TM4 cells in a certain concentration range,which was associated with the activation of ERK signaling pathway.
