神经肽Y干预对海人酸致痫大鼠海马组织神经肽Y表达的影响
The influence of neuropeptide Y intervention on expression of neuropeptide Y in hippocampus for kainic acid-kindled rat

Abstract
Objective To investigate the influence of neuropeptide Y (NPY) intervention on expression of neuropeptide Y in hippocampus for kainic acid-kindled rat. Method 36 male Wistar rats were evenly divided into normal saline group (NS group), kainic acid-kindled rats group (KA group) and NPY group. The kainic acid were injected into the hippocampal CA3 region of the rats in KA group and NPY group to make the chronic epilepsy model. While the rats of NPY group received gene transfection surgery after successful modeling. About 10 μl rAAV2/1-NPY-EGFP (5×1011 μg/ml) were injected into the hippocampal CA3 region of the rats in NPY group by stereotaxic instrument. Equivalent physiological saline were injected into the hippocampal CA3 region of the rats in NS group. The animals were sacrificed and tissues were stored in liquid nitrogen at 2 weeks and 4 weeks after transfection, 6 rats were selected at each time points. The expression level of NPY was detected by immunohistochemistry. On the other hand, positive rate of NPY and staining intensity of positive cells in the CA3 region of each time point rats were scored by semi-quantitative methods. Finally, the positive intensity of NPY expression were calculated. Result After 2 weeks of gene transfection, the rate of NPY positive cells, the staining intensity of positive cells in hippocampus and immunohistochemical score of NPY group and KA group were higher than those of NS group (P < 0.05), while there had no significant differences between NPY group and KA group (P > 0.05). After 4 weeks of gene transfection, the rate of NPY positive cells, the staining intensity of positive cells in hippocampus and IHS score of KA group were higher than those of the other two groups (P < 0.05), and while there had no significant differences between NPY group and NS group (P > 0.05). Conclusion The expression of endogenous NPY in epileptic seizures is significantly increased. It suggests that NPY is involved in the process of antiepileptic and the exogenous NPY has an inhibitory effect on epilepsy.关键词
