Abstract
Objective To investigate the effect of tea polyphenols (TPs) on invasive ability of human prostate cancer PC-3M cells.Methods Human prostate cancer PC-3M cells were treated with 40,60,80μg/ml tea polyphenol for 24h or 48h in vitro.The invasive ability of PC-3M cells was determined by Transwell assay,and the expression of MMP-2,TIMP-2 mRNA and protein was detected by RT-PCR and Western blot,respectively.Results Transwell assay showed that the invasive ability of PC-3M cells was decreased after treatment with tea polyphenol.The expression of MMP-2 mRNA and protein in PC-3M cells was increased after treated with 60μg/ml and 80μg/ml for 24h or 48h were significantly decreased (P<0.05),while the expression of TIMP-2 mRNA and protein was increased (P<0.05) in a dose-dependent and time-dependent manner (P<0.05).Conclusion Tea polyphenols can reduce the invasive ability of human prostate cancer PC-3M cells in vitro,which may be related to the down-regulation of MMP-2 and up-regulation of TIMP-2 gene expression.关键词
