Abstract
Objective:To identify Ziziphi Spinosae Semen and its adulterants using the psbA-trnH sequence.Methods:Genomic DNAs were extracted from Ziziphi Spinosae Semen and its adulterants successfully.The psbA-trnH sequences were amplified and sequenced,and the final sequences were assembled.Multiple sequence alignment was analyzed.The genetic distances were calculated by kimura 2-parameter(K2P) model and the Neighbor-Joining(NJ) phylogenetic tree was constructed.Results:The aligned length of the psbA-trnH sequence of Ziziphi Spinosae Semen was 336 bp.The intraspecific genetic distance of Ziziphus jujuba var.spinosa was 0.017,which was smaller than the interspecific genetic distance of Ziziphus jujuba var.spinosa and its adulterants.The NJ tree indicated that Ziziphi Spinosae Semen distinguished from its adulterants obviously.Conclusion:The psbA-trnH sequence can be used to distinguish medicinal and edible Ziziphi Spinosae Semen from its adulterants accurately and stably.关键词
