Content Determination of Saikosaponins a and d in Bupleurum chinese DC.f.vanheuckii (Muell.-Arg.) Shanet Y.Li. by HPLC Method

Abstract
[Objective]The research aimed to provide scientific basis for evaluating the quality of Bupleurum chinese through establishing HPLC method to determine and study the content of saikosaponins a and d in it.[Method]Thermo ODS-2 HYPERIL(250 mm ×4.6 mm,5 μm)column was used. Mobile phase was acetonitrile-water with gradient elution at a flow rate of 1.0 mL/min. Column temperature was 25 ℃ and the detection wave length was 210 nm.[Result]Methodology examination indicated that the linear relation between peak area integral value and saikosaponins a and d sample were good. RSD in precision experiment was 0.67% and 0.75%, respectively. Meanwhile, the respective recovery coefficient was 99.4% and 98.7%,RSD was 1.53% and 1.76%,respectively. There was only a little difference in the content of saikosaponins a and d that exist in Bupleurum chinese from different origins. The total content was between 0.772% and 1.079%.[Conclusion]This method was stable, simple and with good reproducibility, which can be used for the quality control of Bupleurum chinese. Saikosaponins a and d content in Bupleurum chinese is found higher than the standard content of which in Bupleurum chinense according to Chinese Pharmacopoeia, so Bupleurum chinese can be used as a new resource of bupleurum.关键词
