摘要

Abstract

Objective To investigate the effect of high concentration glucose on the proliferation of bladder cancer T24 cells and to explore its possible mechanism.Methods Bladder cancer T24 cells were cultured in DMEM containing 10% fetal bovine serum,which were divided into five groups: blank group(5.5 mmol/L glucose),control group(5.5 mmol/L glucose+25 mmol/L mannitol),glucose groups(10,20,30 mmol/L glucose).Its influence on the cell proliferation was detected by MTT assay and the clonogenic experiment.The expression of β-catenin mRNA levels was determined by real-time quantitative PCR.Western blot assay was used to detect β-catenin protein expression.Results Compared with the blank and control group,MTT assay and the clonogenic experimen showed that high glucose significantly enhanced the proliferation of T24 cells.The results of Western blot assay showed that high glucose promoted the expression of β-catenin protein in T24 cells.Real-time quantitative PCR results showed that high glucose promote mRNA of β-catenin gene in T24 cells.Conclusion High concentration of glucose significantly promoted the proliferation of bladder cancer T24 cells and promoted the expression of β-catenin protein and its mRNA in T24 cells.The mechanism underlying may be related to the up-regulation of β-catenin expression.
