白花蛇舌草对人前列腺癌DU145细胞增殖和凋亡的影响
The Effect of Hedyotis Diffusa Extract on Proliferation and Apoptosis of Prostate Cancer Cell Line DU145
摘要
Abstract
Objective:To investigate the effect of Hedyotis diffusa extract on proliferation and apoptosis of prostate cancer cell line DU145 and its possible mechanisms.Methods:Prostate cancer cell line DU145 were treated with different concentration of Hedyotis diffusa extract(0.25 g · L-1,0.5 g · L-1,1 g · L-1)for 24 h,48 h,72 h,CCK-8 assay was used to detect the proliferation of DU145 cell.Flow cytometry was performed to detect cell apoptosis;RT-PCR was carried out to detect the mRNA expression of COX-2 and PCNA;Western bloting was used to examine the protein expression of Bcl-2,Bax,Caspase3 and CyclinD1.Results:The inhibited effect of Hedyotis diffusa extract in DU145 proliferation exhibited dose and time dependent manner.The concentration of Hedyotis diffusa extract treated longer than 48 h could inhibit the proliferation of DU145 cells,the COX-2 mRNA,PCNA mRNA and Bcl-2,Cyclin D1 expression of DU145 cells was significantly decreased,Bax and Caspase3 expression significantly increased,the change degree was in a dose-dependent manner.Conclusion:Hedyotis diffusa extract can inhibit the proliferation of DU145 cells and induce the apoptosis of DU145 cells,which may occour though change the expression of cell cycles and apoptosis-related protein.关键词
