Abstract

In order to investigate the genetic diversity and provide theoretical basis for rational utilization of Zhongdian yak,this study analyzes the complete sequence of 15 individuals' ND6 gene.We analyzed the diversity of nucleotide,composition and diversity of haploid type by MEGA5.0,DNASP5.0,Clustal X1.83,etc.The results showed that (1) the length of the ND6 gene of Zhongdian yak were 528 bp,with nucleotide frequencies of 20.8%,42.2%,7.6% and 29.4% for T,A,G,and C respectively,there are some bias of base composition.The length of the COⅢ gene of Zhongdian yak were 781 bp,with nucleotide frequencies of 29.2%,26.1%,15.2%,29.5% for T,A,G,and C respectively.(2)There are no haploid and mutant sites in 15 individuals of Zhongdian yak,showing that the genetic diversity of Zhongdian yak's ND6 gene is relatively poor;Three haplotypes were identified of 15 individuals,with four polymorphic sites,including two conversions,two transversions;(3) In evolutionary tree,Zhongdian yak and Maiwa yak clustered first,then gathered with Bos mutusc.It means there are close genetic relationship between Zhongdian yak and Bos mutusc;Known from the analysis of genetic distance,Zhongdian yak has the minimum distance with Bos mutusc,and the maximum distance with Bubalus bubalis.The result supported the point that Bos grunnuens and Bos mutus are classified as poephagus of Bovinae.
