川楝子水提物对肺癌A549细胞增殖的抑制作用
Toosendan aqueous extract inhibits proliferation of lung cancer A549 cells in vitro
摘要
Abstract
Objective:To investigate the effect of Toosendan aqueous extract on human lung cancer cell line A549 cells.Methods:This study was divided into experimental group (concentration of 200,150,100,50μg/ml) and control group.The inhibitory rates of A549 cells effected by Toosendan aqueous extract were detected using MTT assay,and the changes of cell morphology were observed by microscope.The mRNAs expression of of p53,Bax,Bcl-2,Bad and Bcl-xl were detected using quantitative real-time PCR assay.Results:Proliferation of A549 cell was inhibited by Toosendan aqueous extract,and inhibition rates were related with time and the extract concentration in MTT assay.Compared with the control group,expression of P53,Bax and Bad mRNA increased,and expression of Bcl-2 and Bcl-xl mRNAs decreased in the experimental group.Conclusion:Toosendan aqueous extract may inhibit proliferation of A549 cells via activation of P53 gene and mitochondrial apoptotic pathway.关键词
