摘要

Abstract

Objective To explore the possible role of IL-1β,TNF-α and IL-10 in the pathogenesis of epilepsy, and investigate the effect of gastrodin on IL-1β,TNF-α and IL-10 in hippocampus neurons from epileptic rats.Methods Forty-five adult male Wistar rats were randomly divided into normal control group (NC group), the epilepsy group (EP group) and gastrodin group (GE group).EP group and the GE group injection subthreshold doses of Pentylenetetrazol by intraperitoneal once a day,until them reache the ignited standards.GE group injection gastrodin by intraperitoneal.Observed and recorded the changes of rats''s behavior.IL-1β,TNF-α and IL-10 levels change in hippocampal neurons were detected and compared by immunohistochemistry.Results Compared gastrodin group with epilepsy group the incubation period of onset was significantly prolonged, the duration is shorter、attack strength is lower (P<0.05).Compared with normal control group, IL-1β,TNF-α levels in hippocampal neurons of epilepsy group significantly increased(P<0.01), and IL-10 levels decreased significantly(P<0.01).Compared with epilepsy group,IL-1β,TNF-α levels in hippocampal neurons of gastrodin group decreased significantly (P<0.01),and IL-10 levels increased significantly (P<0.01).Conclusions IL-1β、TNF-α can promote the occurrence and development of epilepsy, antiepileptic and IL-10 has the effect of antiepileptic.The gastrodin play an antiepileptic effect by lower IL-1β,TNF-α and increased IL-10 levels.
