Abstract

Objective To investigate the effect of icariin (ICA) in inhibiting inflammatory resoponse and improving learning and memory abilities in ICR mice with lipopolysaccharide (LPS) induced Alzheimer's disease (AD).Methods A total of 18 ovariectomized mice were randomly divided into control group,LPS group,and ICA+ LPS group,with 6 mice in each group.The mice in the control group were given dimethyl sulfoxide by gavage.The mice in the LPS group were given injection of LPS in the lateral ventricle to establish a mouse model of AD,followed by dimethyl sulfoxide by gavage.The mice in the ICA+LPS group were given injection of LPS in the lateral ventricle and then continuously given ICA by gavage.The course of treatment was 12 days.The water maze test was used to test learning and memory abilities,real-time reverse transcription-polymerase chain reaction was used to measure the mRNA expression of inducible nitric oxide synthase (iNOS) and tumor necrosis factor-α (TNF α).Results Compared with the control group,the LPS group had significant reductions in the proportion of time spent in the target quadrant and number of platform crossings and significant increases in the mRNA expression of iNOS and TNF-α in the hippocam pus.Compared with the LPS group,the ICA+LPS group had significantly less injury induced by LPS,significant increases in the proportion of time spent in the target quadrant and number of platform crossings,and significant reductions in the mRNA expression of iNOS and TNF-α (F=9.214-57.090,q-4.114-13.400,P<0.05).Conclusion In ICR mice with LPS-induced AD and inflammation,ICA can inhibit the mRNA expression of iNOS and TNF-α in the hippocampus and improve their learning and memory abilities.
