Abstract

Objective To examine the effect of ferric ammonium citrate (FAC) on the release of pro-inflammatory cytokines by C6 glioma cells exposed to extracellular α-synuclein (α-syn).Methods C6 cells were stimulated with α-syn,FAC,α-syn+ FAC,or serum-free medium (control).The mRNA expression of intracellular interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) was measured using real-time PCR.Results The IL 1β and TNF-α expression in the α-syn group was significantly higher than that in the control group (F=5.43-67.19,q=4.07-16.25,P<0.05).On the other hand,while there were no significant differences in IL 1β and TNF-α expression between the 10 μmol/L FAC group and the control group (P>0.05),IL 1β and TNF-α expression was significantly enhanced in the 100 μmol/L FAC group compared with the control group (q =14.48 4.14,P <0.05).However,there were no significant differences in IL-1β and TNF-α expression between the α-syn+ FAC group and α-syn group (P>0.05).Conclusion Extracellular α-syn can stimulate C6 cells to release pro-inflammatory cytokines,which is not affected by the increase in iron levels.
