Abstract

Objective To investigate the effect of 5-hydroxytryptamine (5-HT) on spontaneous discharge in globus pallidus neurons in normal mice.Methods Eight mice were randomly divided into two groups,with four mice in each group.The mice in control group were given micro-pressure injection of normal saline,and those in the experimental group were given micropressure injection of 5-HT.Extracellular electrophysiological recording was performed to observe the changes in discharge frequency,coefficient of variation (CV) of spontaneous discharge interval,and fano factor (FF) of globus pallidus neurons before and after injection.Results A total of 15 globus pallidus neurons with discharges were recorded in the experimental group,with a meandischarge frequency of (6.16±0.93) Hz and a CV of 0.39±0.11;after micro-pressure injection of 5-HT,13 globus pallidus neurons had a significant increase in the frequency of spontaneous discharge (t =2.95,P<0.05).Compared with the control group,the experimental group had a significant increase in discharge frequency after injection (Z-2.94,P <0.01).In the experimental group,there were two discharge patterns of globus pallidus neurons;5 globus pallidus neurons (33.33%) had regular discharge and 10 (66.67%) had irregular discharge.Micro-pressure injection of 5-HT or normal saline had no significant influence on CV or FF (P>0.05).Conclusion 5-HT can excite globus pallidus neurons in normal mice.
