脐带间充质干细胞对衰老心肌细胞炎症因子表达影响
EFFECT OF UMBILICAL CORD MESENCHYMAL STEM CELLS ON EXPRESSION OF INFLAMMATORY FACTORS IN SENESCENT CARDIOMYOCYTES

Abstract
Objective To investigate the effect of human umbilical cord mesenchymal stem cells (hUC-MSCs) on the expression of inflammatory factors in senescent cardiomyocytes induced by hydrogen peroxide (H2 O2).Methods Senescence of H9c2 cardiomyocytes was induced by 100 μmol/L H2 O2,and DAPI fluorescent staining was used to measure the proportion of cells with senescence-associated heterochromatin foci (SAHF),in order to determine whether the senescence model was successfully established.Transwell chambers were used to establish a co-culture system of H9c2 cardiomyocytes and hUC-MSCs;DAPI fluorescent staining was used to investigate the influence of hUC-MSC culture on the proportion of SAHF-positive H9c2 cardiomyocytes,and ELISA was used to measure the expression of tumor necrosis factor-α (TNF α),interleukin-1β (IL-1β),and interleukin-10 (IL-10) in supernatant.Results DAPI fluorescent staining showed that compared with the control group,the H2O2 group had a significant increase in the proportion of SAHF-positive cells (F=22.658,q=9.562,P<0.05),suggesting that the model of cardiomyocyte senescence was established successfully.Compared with the H2 O2 group,the H2 O2 +hUC-MSCs group had a significant reduction in the proportion of SAHF positive cells (q =7.092,P<0.05).ELISA results suggested that compared with the H2 O2 group,the H 2O2 +hUC-MSCs group had significant reductions in the expression of proinflammatory factors TNF-α (F =7.338,q=4.334,P<0.05) and IL-1β (F=13.965,q=6.144,P<0.05) in supernatant,as well as a significant increase in the expression of the anti-inflammatory factor IL-10 (F=17.995,q=6.405,P<0.05).Conclusion hUC-MSCs have a protective effect against the senescence of H9c2 cardiomyocytes induced by H2 O2,possibly by reducing the expression of proinflammatory factors and increasing the expression of anti-inflammatory factors.关键词
