牙龈间充质干细胞的培养与鉴定
CULTURE AND IDENTIFICATION OF GINGIVAL MESENCHYMAL STEM CELLS

Abstract
Objective To investigate the isolation and culture of gingival mesenchymal stem cells (GMSCs),and to provide a basis for further research on the therapeutic effect of GMSCs on periodontitis.Methods The enzyme digestion method was used to isolate GMSCs from gingival tissues,and plating efficiency,adipogenic and osteogenic differentiation,and flow cytometry were used to measure the abilities of proliferation and multi-directional differentiation and expression levels of markers.Results GMSCs adhered to the wall and had a similar spindle shape as fibroblasts and marked colonies.GMSCs produced mineralized nodules and lipid droplets under specific induction conditions.The stem cell markers CD73,CD90,and CD105 had an expression rate of ＞95％,and the hematopoietic stem cell marker CD45 had an expression rate of ＜2％.Conclusion GMSCs isolated by the enzyme digestion method have the abilities of self-renewal and multi-directional differentiation and can express stem cell markers,which lays a foundation for future experiments.关键词
