DHF对过氧化氢损伤PC12细胞的保护作用及机制
MECHANISM OF ACTION OF 7,8-DIHYDROXYFLAVONE IN PROTECTING PC12 CELLS AGAINST HYDROGEN PEROXIDE-INDUCED DAMAGE

Abstract
Objective To examine the protective effect of 7,8-dihydroxyflavone (DHF) against hydrogen peroxidc (H2O2) induced PC12 cell damage and its underlying mechanism of action.Methods PC12 cells were incubated with different concentrations of H2O2(300-800 μmol/L) for 24 h to observe the toxic effect of H2O2 on PC12 cells.The cells were then divided into six groups,namely control group,H2O2 group (cells were incubated with 600 μmol/L H2O2 for 24 h),DHF plus H2O2 group (cells were pretreated with 25 μmol/L DHF for 6 h,then exposed to 600 μmol/L H2O2 for 24 h),zinc protoporphyrin (ZnPP)-pretreated group (cells were pretreated with 5 μmol/L ZnPP for 30 min and then exposed to DHF plus H2O2),DHF group (cells were incubated with 25 μmol/L DHF only for 24 h),and ZnPP group (cells were incubated with 5 tμmol/L ZnPP only for 24 h),in order to observe the protective effect of DHF against H2O2 induced PC12 cell damage.Cell viability in each group following treatment was measured using the MTT assay.Results Cell viability was significantly reduced with H2O2 treatment in a dose-dependent manner (F=34.06,q=4.48-15.61,P<0.05).Pretreatment with DHF elevated the viability of H2O2-damaged PC12 cells,and this effect could be blocked by the heme oxygenase-1 (HO-1) inhibitor ZnPP (F =18.10,q =2.37-10.56,P <0.05).Conclusion DHF can significantly protect PC12 cells from H2O2-induced damage,and this protective effect may be mediated by the upregulation of HO-1 expression.关键词
