Abstract

Objective To investigate the expression changes of serum miRNA-34 and miRNA-500 in anti-SSA posi-tive patients with primary Sjogren's Syndrome (pSS)and to analyze their clinical value in diagnosis of anti-SSA positive pSS. Methods Serum samples were obtained from 75 anti-SSA positive patients with pSS (observation group)and 84 an-ti-SSA positive patients without pSS (control group). miRNA microarray was used to determine the alteration of miNRA ex-pression profiles between the above two groups and the results were validated by RT-PCR. The value of miRNAs with abnor-mal expression in diagnosis of anti-SSA positive pSS were analyzed by using statistical methods. Results A total of 10 miRNAs showed altered expression in anti-SSA positive pSS patients,including 6 up-regulated miRNAs and 4 down-regula-ted miRNAs,among which,the most remarkable increase was observed in miR-34 and miR-500 of the observation group. Compared with the control group,the serum miR-34 and miR-500 were significantly increased in the observation group, both P < 0. 001;receiver operating characteristic (ROC)analysis showed that,AUC value of miR-34 in diagnosing anti-SSA positive pSS was 0. 783 (95% CI:0. 710-0. 844,P < 0. 001),and AUC value of miR-500 was 0. 774 (95% CI:0. 701-0. 837,P < 0. 001)in the observation group;AUC value of miR-34 combined with miR-500 in diagnosis of anti-SSA positive pSS was 0. 888 (95% CI:0. 828-0. 932),which was superior to a single detector miR-34 or miR-500 (all P <0. 05). Conclusion The expression of serum miR-34 and miR-500 in anti-SSA positive patients with pSS is increased, and the combined detection of miR-34 and miR-500 is helpful in diagnosis of anti-SSA positive pSS.
