摘要

Abstract

Objective To investigate the protective effect of extraction of Eucommia ulmoides oliv (eucommin) on the function of human sperm membrane under oxidative injury.Methods The normal sperms were isolated with percoll gradient centrifugation, and the reactive oxygen species (ROS) were produced with hypoxanthine-xanzine xanzine (HX-XO) system.The human sperms suspension in ROS environment was treated with vitamin C (0.15mg/ml) or different concentrations of eucommin (150, 300 and 600mg/L).The malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of sperm membranes were detected.The function of sperm membrane was analyzed by sperm hypotonic expansion test and the rate of intact acrosome was detected.Results Compared with oxidative model group, SOD activity of sperm suspension was improved,and the content of MDA was reduced in vitamin C group and eucommin groups ( P <0.01);the effect of high-dose eucommin group was more marked than that of vitamin C group ( P <0.01).There was significant difference in the rate of intact acrosome and sperm hypotonic expansion test between model group and eucommin groups ( P <0.01), and the high-dose eucommin group showed better result than other groups ( P <0.01).Conclusion Eucommin has a protective effect on human sperm membrane function from oxidative injury.
