摘要
Abstract
Objective:To investigate the influence of the change of β-catenin induced by O-GLcNAcylation in ovarian cancer cells.Methods:SKOV3 cell was transfected with lentivirus to bulid O-GLcNAc lower expression model.A2780 was treated with PUGNAc,a inhibitor of O-GLcNAc hydrolase (OGA),to bulid O-GLcNAc higher expression model.Fowcytometry technology was used to test cell cycle.MTT was used to test cell proliferation.Western blot was used to test β-catenin expression in different cell models.CO-IP was used to test the glycosylation level of β-catenin in different cell models.Results:Cell proliferation was influenced by the level of O-GLcNAcylation.The levels of O-GLcNAcylation mediated the expression of β-catenin in ovarian cancer cell.Conclusion:High levels of O-GLcNAcylation can enhance the expression of β-catenin,and thus influence cell proliferation in ovarian cancer cell.关键词
