The expression and significant of autophagy related genes BECN1,LC3B and mTOR in the progression of cervical squamous lesions

Abstract
Objective:To detect the expression of BECN1,microtubule-associated protein 1 light chain 3 and mammalian target of rapamycin in cervical squamous lesions and to explore their clinical significance.Methods:The protein expressions of BECN1,LC3B and mTOR were evaluated by immunohistochemistry methods in cervical squamous lesions.Detecting the mRNA transcription level by using fluorescence in situ hybridization.In addition,normal cervical tissue was used as control group.Results:In cervical squamous cell carcinoma (SCC) and high grade squamous intraepithelial lesions (HSIL)group,the protein and mRNA expression levels of BECN1 and LC3B were significantly lower than that in control group (P<0.01),while the protein and mRNA expression levels of mTOR were significantly higher than that in control group (P<0.01).In the low grade squamous intraepithelial lesion (LSIL) group,the protein expression of BECN1 and LC3B were significantly lower than that in control group (P<0.01),the mRNA expression of LC3B was significantly lower than in control group (P<0.01),while the mRNA expression of mTOR was significantly higher than that in control group (P<0.01).In cervical squamous cell carcinoma group,the expression of BECN1 related with clinical stage and lymphatic metastasis.LC3B related with differentiation and mTOR related with lymphatic metastasis.The expression of LC3B was positively correlated with BECN1 and negatively correlated with mTOR in cervical squamous cell carcinoma.Conclusion:Autophagy related genes are associated with the severity of cervical squamous lesion,suggesting that abnormal expression of BECN1,LC3B and mTOR may lead to the deterioration of the lesions.关键词
