摘要

Abstract

Objective:To establish a stable cell model of intrauterine adhesions and to detect the expression of forkhead box F2(FoxF2).Methods:Human endometrial stromal cells(HESCs) cultured by primary method were identified by immunofluorescence staining.Applied 0,2.5,5,10ng/ml transforming growth factor β1(TGF-β1) to HESCs for 48 hours and then detected the mRNA and protein expressions of α-smooth muscle actin(α-SMA),collagen I(COLI) and FoxF2 by PCR and Western blot.Applied 10ng/ml TGF-β1 to HESCs for 12,24,48,72hours and then detected the mRNA and protein expressions of α-SMA,COLI and FoxF2 by same methods.Results:The mRNA and protein expressions of α-SMA,COLI and FoxF2 were increased gradually in the groups of 2.5,5ng/ml and 10ng/ml compared with the 0ng/ml group.As the effect time of 10ng/ml TGF-β1 was prolonged,the mRNA and protein expressions of α-SMA,COLI,and FoxF2 were also increased gradually in the groups of 24,48 and 72hours compared with 12 hours group.Conclusion:TGF-β1 can induce the fibrosis of HESCs,thus the cell model of intrauterine adhesions was established.The expressions of FoxF2 increased significantly in intrauterine adhesions.
