欧洲型PRRSV辽宁株LNEU12的分离及其ORF5的序列分析
Isolation and ORF5 sequence analysis of EU-PRRSV LNEU12 strain from Liaoning Province
摘要
Abstract
A virus strain named LNEU12 was isolated by Marc-145 cells from the serum of a dead pig suspicious of porcine reproductive and respiratory syndrome virus (PRRSV) infection,and this viral strain could induce regular cytopathogenic effects (CPE) on Marc-145 cells after five passages.The virus had a mean diameter of about 55 nm,as measured by electron microscope.ORF5 gene of the virus isolated was amplified by RT-PCR and sequenced after being cloned directly into the pMD18-T.Sequence analysis showed that the ORF5 gene of the virus shared 87.1％ and 87.6％ of sequence identities with that of Lelystad virus and Amervac strain,and 83.5％ to 94.2％ identities with domestic strains of EU-PRRSV.Compared with LV strains,22 amino acids of GP5 showed the mutation including three potential N-glycosylation sites.The phylogenetic tree revealed LNEU12 had closer relationship with BJEU06-1and NVDC-NM1-2011 strains than that of VR2332 strain.It is inferred that the LNEU12 strains was the European type PRRSV.关键词
