摘要

Abstract

AIM:To explore the effect of oligomeric proantho cyanidins (OPC) on learning and memory and cAMP-response element binding protein (CREB) expression of Alzheimer disease (AD) rats.METHODS:Forty elder rats were randomly divided into four groups,i.e.the blank control group,model group,OPC high dose group and OPC low dose group.Except the normal group,rats of the other groups were cultivated into AD models by basal forebrain-injure.Morris water maze was applied to detect the learning and memory ability.Double anti-body sandwich method was used to test the activity of AchE and CHAT;Western blot was used to detect the expression of CREB protein.RESULTS:Compared with blank control group,the escape latency and distance of search were significantly longer;the search time and exploring ratio were significantly lower;the activity of AchE was significantly higher;the activity of ChAT and the expression of CREB were significantly lower.Compared with model group,OPC high dose group and OPC low dose group presented shorter escape latency and search distance,combined with longer exploring time and higher exploring ratio.The activity of AchE was significantly decreased while the expression of ChAT and CREB protein were significantly increased (P < 0.05,P < 0.01).CONCLUSION:OPC can improve the learning and memory of AD rats by decreasing AchE and increasing ChAT and CREB expression.
