摘要

Abstract

Objective To investigate the effect of curcumin on proliferation and metastasis of oral squamous cell carcinoma HN4 cell line and the molecular mechanism. Methods Oral squamous cell carcinoma HN4 cells were cultured and treated with 0-60 μmol/L curcumin, and MTT was used to assess the cell viability after 24 hours. Wound scratch assay was performed to detect the invasion of the HN4 cells. Cell cycle and apoptosis were detected by flow cytometry. The protein expression levels of p53, E-cadherin and Snail were detected by Western blot. Results MTT assay revealed that curcumin significantly inhibited the growth of the cancer cells at the experimental concentrations, the effect was more obvious at the concentration of 20 μmol/L. With the increase of curcumin concentration, the effect on cell apoptosis was getting more significant. The HN4 cells were arrested at G0/G1 phase after treatment. With the increase of curcumin concentration, the expression of Snail protein decreased, and the expressions of p53 and E-cadherin increased. Conclusions Curcumin can significantly inhibit the growth of oral squamous cell carcinoma HN4 cell line, and the mechanism may be up-regulation of p53 expression.
