Abstract

Objective To observe the proliferation and invasion abilities of ten-eleven translocation 1 (TET1) gene knock-out Hela cervical cancer cells.Methods We constructed the TET1 knock-out Hela cells by CRISPR/Cas9 technology,which were defined as the observation group.At the same time,we defined wild Hela cells as the control group.We observed the proliferation ability of Hela cells in the observation and control group by the real-time cellular analysis (RTCA),and calculated the cell index (CI) values in each group at 90 h.We observed the invasion ability of Hela cells in each group by Transwell assay.Results The CI value of the observation group was 1.04 ± 0.86,which was higher than that of the control group (i.47 ± 1.01) (P <0.05).At 24 h,the transmembrane cells in the observation group and the control group were 158 ± 6 and 109 ± 4,respectively;at 48 h,the of transmembrane cells in each group were 206 ± 12 and 142 ± 7,respectively;significant difference was found between these two groups (P < 0.05).Conclusion The proliferation and invasion abilities of TET1 gene knock-out Hela cells become stronger.
