Abstract

To explore the extraction method of trace components in fresh raw silk, this paper is intended to study the effect of extraction solvents (ethanol/water solution) of different mass fractions and extraction conditions (temperature, time and liquor ratio) on extraction efficiency of polyphenols trace components in fresh raw silk via high performance liquid chromatography (HPLC), figure out the optimum extraction conditions via orthogonal test.The results indicate that the HPLC chromatograms of extracts from fresh raw silk with different solvents significantly differ.Specifically, with the mass fraction of ethanol is in negative correlation with both the intensity of chromatographic peaks and the content of components.All the extraction conditions have impact on the content of components having retained for 10 to 16min, and the impact of temperature is most significant.Meanwhile, researches show that pure water is the best extraction solvent of polyphenols trace components in fresh raw silk, of which the optimum extraction conditions are: temperature of 40℃, duration of 90min, and liquor ratio of 1:25 (g/mL).
