三阴性乳腺癌患者miR-155表达水平及其对预后的预测价值研究
Expression and Prognostic Significance of miR-155 in Human Triple-negative Breast Cancer

Abstract
Objective To investigate the expression and prognostic significance of miR-155 in patients with triple-negative breast cancer(TNBC).Methods Formalin-fixed and paraffin-embedded tumor samples of 72 patients with TNBC who received mastectomy at the First Affiliated Hospital of Soochow University between June 2006 and December 2008 were obtained from the Department of Pathology of the hospital.The data of pathology and follow-up were collected.The real-time quantitative polymerase chain reaction(RTQ-PCR) was employed to detect miR-155 levels in breast cancer tissues.Moreover,two-step immunohistochemical staining was used to test the expressions of Ki-67 in the same specimen.The predictive accuracy of miR-155 and Ki-67 for recurrent TNBC was assessed by a receiver operating characteristic(ROC) test and evaluated by area under curve(AUC).Results The median miR-155 expression level was 0.899(1.236)in 72 patients with TNBC.With 0.899 as the cutoff value of miR-155,patients were divided into two groups as miR-155 low expression and miR-155 high expression.Analysis with chi-square test revealed that miR-155 expression was not related to patient′s age,tumor grading,clinical stage of tumor,Ki-67 expression,menopause status,lymph node metastasis(P>0.05).Recurrence of TNBC occurred in 25 cases(34.7%) three years after the surgery.Significantly higher miR-155 expression was found in the recurrent group than in the non-recurrent group with TNBC(P<0.05).AUC scores of ROC were 0.777 for miR-155,0.636 for Ki-67,and 0.829 for miR-155 combined with Ki-67,respectively.Conclusion Patients with recurrent TNBC have higher expression of miR-155 than those without.miR-155 may serve as a useful prognostic biomarker of postoperative TNBC recurrence.关键词
