乌珠穆沁羊不同组织基因组DNA甲基化状态的SAP分析
Analysis of DNA Methylation in Different Ujumqin Sheep Tissues with MSAP

Abstract
The CCGG sites in genomes were detected among heart,liver,spleen,lung,kidney,stomach and longissimus muscle of 5 Ujumqin sheep for 10 pairs of selective primers by methylation sensitive amplified polymorphism (MSAP) method.A total of 1249 DNA fragments were detected by the polyacrylamide gel electrophoresis,including 154 type Ⅱ and 231 type Ⅲ fragments.Through calculation the methylation rates of different tissues for Ujumqin sheep,the average methylation rate of heart,liver,spleen,lung,kidney,stomach and longissimus muscle was 37.58%,39.88%,50.63%,25.82%,20.53%,28.19%,19.21% respectively.For the all detected tissues,the spleen tissue methylation degree was The highest and the muscle tissue was the lowest.This study laid the foundation for further understanding of the role of DNA methylation regulatory effect for the sheep tissue differentiation.关键词
