不同频率电针对神经病理性疼痛诱发焦虑大鼠ACC区磷酸化ERK水平的影响
Effects of Electroacupuncture with Different Frequencies on p-ERK Levels in ACC of Neuropathic Pain Induced Anxiety Rats

Abstract
Objective To observe the effects of electroacupuncture (EA) with different frequencies on the expression of phosphorylated extracellular signal-regulated kinase (p-ERK) in anterior cingulate cortex (ACC) of rats in neuropathic pain induced anxiety rats,and to study the optimal frequency of EA for neuropathic pain induced anxiety and its possible mechanism.Methods Totally 48 SD rats were randomly divided into 6 groups,i.e.,the normal group,the sham-operation group,the model group,2 Hz EA group,15 Hz EA group,and 100 Hz EA group,8 in each group.Spinal nerve ligation (SNL) model was established in 32 rats (L5).Of them,lumbar 5 spinal nerve was separated in the sham-operation group,but without ligation.Rats in 2 Hz,15 Hz,and 100 Hz EA group were respectively needled at bilateral Huantiao (GB30) and Yanglingquan (GB34) with 2 Hz,15 Hz,and 100 Hz,once every other day,30 min each time for a total of 13 times.Paw withdrawal thresholds (PWT) was detected as mechanical pain threshold using Dynamic Plantar Aesthesiometer.Elevated zero maze test (EZMT) was used to evaluate the level of anxiety of rats.And then Western Blot was used to determine the expression of p-ERK in ACC.Results The PWTs showed,as compared with the sham-operation group,the ipsilateral PWTs were obviously shorter in the model group than in the sham-operation group at day 3 after SNL (P <0.01).The ipsilateral PWTs showed an increasing tendency in rats of 2 Hz,15 Hz,and 100 Hz EA groups at day 8,12,16,20,and 28 after operation,higher than those of the model group at the same time points (all P <0.01).The ipsilateral PWTs in rats of 2 Hz EA group were higher than those of 15 Hz EA group and 100 Hz EA group at the same time points (allP <0.01).Scores in EZMT showed,as compared with the sham-operation group,open arm time (OAT) was reduced more obviously in rats of the model group (P <0.05).Besides,OAT in rats of 15 Hz EA group and 100 Hz EA group were obviously longer than that in rats of the model group (P <0.05).No differences were found in OAT between 2 Hz EA group and the model group (P >0.05).The expression level of p-ERK in ACC showed,the expression of p-ERK in ipsilateral ACC of the model group was obviously higher than that in rats of the sham-operation group (P <0.01).The protein expression of p-ERK in ipsilateral ACC in rats of 2 Hz,15 Hz,and 100 Hz EA groups were obviously lower than that in rats of the model group (P <0.05).The protein expression of pERK in ipsilateral ACC in rats of 100 Hz EA group were obviously lower than that in rats of 2 Hz EA group and 15 Hz EA group (P <0.05,P <0.01).Conclusions The optimal frequencies of EA in intervening neuropathic pain and its induced anxiety were various.2 Hz EA showed better effects on neuropathic pain.But for neuropathic pain induced anxiety,100 Hz EA showed better curative effect,which had close relationship with down-regulating the expression of p-ERK in ACC.关键词
