The Effect of HCMV Infection on the Stemness of Down-regulated ATF5 U251 Cells

Abstract
To study the effect of HCMV infection on the expression of CD133, cell proliferation and clone formation of U251 cells, which were down regulated the ATF5 expression.The changes of IE, CD133 gene and protein expression levels of HCMV (MOI=1) infected U251 cells were detected by fluorescence quantitative PCR and Western-blot methods.The results showed that the expression of CD133 increased significantly with the increase of IE expression in U251 cells.Silencing ATF5 cell line siATF5-U251 was successfully constructed, and HCMV infection reduced the cell proliferation activity, and decreased the colony formation rate.The results of fluorescence quantitative PCR and Western-blot showed that HCMV infection significantly inhibited the expression of CD133 in siATF5-U251 cells.Therefore, inhibiting the expression of ATF5 may be a new target for prevention and treatment of malignant glioma.关键词
