Effects of dental papillae cells on the proliferation of dental pulp stem cells and its mechanism

Abstract
Objective To investigate effects of dental papillae cells on the proliferation of dental pulp stem cells (DPSCs) and its mechanism.Methods DPSCs and dental papillae cells were cultured,the co-cuhured systems of DPSCs and dental papillae cells were established.The intracellular mineralization nodes were observed.The cell cycle of DPSCs was detected by flow cytometry.Finally,when the inhibitor of Notch,PHI,was administrated,the expression of Notch1,Jagged1 mRNA and protein was examined.Results The obvious mineralization nodules were observed in DPSCs after 14 d of co-culture.Flow cytometry results showed that stratified co-culture promoted the proliferation of DPSCs.RTPCR and western blot results showed that the expression of Notch 1,Jagged 1 mRNA and protein in the co-culture group was significantly increased compared with control group.Conclusion Stratified co-culture of DPSCs and dental papillae cells might promote the mineralization and proliferation by activating Notch signal pathway.关键词
